The aim of the study was to assess the effect of Chlamydia trachomatis lipopolysaccharide (LPS) on the fertility and viability of gametes under in vitro conditions. A new method for the preparation of C. trachomatis LPS is proposed, based on affinity chromatography on concanavalin A-Sepharose, which allows to improve the quality of endotoxin, while reducing the toxicity of the method. The spermatozoa of 25 fertile men were incubated for 6 hours with 0.1 μg / ml LPS C. trachomatis (serovar E). The externalization of phosphatidylserine (PS) was determined by staining spermatozoa with conjugated with fluorochrome annexin-V and propidium iodide followed by fluorescence microscopy. The ability of C. trachomatis LPS to decrease the motility of gametes and induce externalization of PS is revealed, which is an early sign of apoptosis. LPS, as one of the pathogenicity factors of C. trachomatis, is able to exert a cytotoxic effect on human spermatozoa, inducing apoptosis-associated structural and biochemical changes in the sperm membrane and re-ducing gamete fertility.
