COLLAGENASE AND HYALURONIDASE AS TEST-OBJECTS FOR THE PRIMARY SCREENING OF THE COMPAUNDS HAVING ANTIARTHRITIS ACTIVITY

An original in vitro test-system for the primary detection of biologically active compounds and drugs with antiarthritic and antiinflammatory action has been developed. As test objects the hydrolytic enzymes of connective tissue ─ collagenase and hyaluronidase were used. To prove the adequacy of the test- objects, nonsteroidal anti-inflammatory drugs with proven antiarthritic activity were tested. The ability of test compounds to inhibit these enzymes in an indication of BAS antiarthritic properties.The techniques to determine the rate of collagenase and hyaluronidase activity in the presence of the test BAS and the drug in vitro conditions were developed. Studying of the kinetic parameters of enzymatic reactions provides additional information on both the structure and function of enzymes and how LP or BAS interact with them.
