DETERMINATION OF MITOMYCIN-C AND SOME OTHER NITROGEN-CONTAINING HETEROCYCLIC COMPOUNDS BY HPLC METHOD WITH AMPEROMETRIC DETECTOR

High performance liquid chromatography with amperometric detector (HPLC-AD) currently used for the analysis of biologically
active compounds, as well as a number of drugs. Amperometric detector is specific selective for certain classes of compounds that has
a low detection limit and is used to quantify the amount of antioxidants in the analyzed samples of foods and beverages.
The aim of this study was to investigate the possibility of using amperometric detector for qualitative and quantitative determination
of mitomycin-C and some benzimidazole derivatives in various objects. Chromatographic investigation was carried out by
reversed phase high performance liquid chromatography (RP-HPLC). The experiment was performed by liquid chromatograph "Cvet
Yauza" using amperometric detection (AD). It is found that the minimum concentration of mitomycin-C detectable by amperometric
detector is 32 μg/l.
It is shown that amperometric detector is sensitive to even minor changes in the structure of the benzimidazole. Adding
methanol to the water-acetonitrile solution increases the sensitivity of amperometric detector for all investigated benzimidazole
derivatives.
