PROTEOLYTIC ACTIVITY COMPARATIVE EVALUATION OF BACTERIA AND MICROMYCETES

Obtaining various biologically active substances using of biotechnological approaches is one of perspective directions of modern scientific research. In recent years the list of such substances, including proteases with different substrate specificity was significantly expanded. It should be noted, the different collagenases and keratinases with their ability to hydrolyze collagen and keratin are among these enzymes. These enzymes are promising for use in medicine, cosmetics, various industries and agriculture. Most of proteases are produced abroad. Previously it was shown that some micromycetes from FGBNU VILAR biocollection secrete proteolytic enzymes to the culture media. This investigation is devoted to the proteolytic activity comparative study of some bacteria and filamentous fungi to as-sess the prospects for their further use as producers. In the present work, we first estimated the ability of 3 species of bacteria and 2 species of micromycetes from the collection of FGBNU VILAR to hydrolyze various substrates containing collagen or keratin by using the screening method during surface cultivation of microorganisms. It was shown that all investigated cultures were grown and formed zones of lysis in media containing collagen, gelatin and liquid keratin that indicates the suitability for their further study as potential producers of collagenases and keratinases. Fungi as well as bac-teria less actively grew and formed zones of lysis in media containing keratin, in comparison with media containing collagen or gelatin. The growth on media containing keratin of the hair was minimal. Aspergillus rubber was the only culture which formed zones of lysis on this medium. Indexes of lysis in bacteria and fungi comparative analysis was shown that the fungi proteolytic potential was higher. However bacteria had the high rate of biomass accumulation, which can provide their advantages use as producers.
