ТРАНСПЛАНТАЦИЯ МИТОХОНДРИЙ ДЛЯ ТЕРАПИИ БОЛЕЗНИ АЛЬЦГЕЙМЕРА (ОБЗОР)

Болезнь Альцгеймера (БА) является наиболее распространенной формой деменции, которая поражает преимущественно пожилых людей и чаще всего начинается с едва заметного ухудшения памяти с последующим прогрессирующим нарушением поведенческих и когнитивных функций. Несмотря на то, что основными патологическими признаками БА считаются внеклеточные отложения бета-амилоида в виде амилоидных бляшек и внутриклеточное накопление гиперфосфорилированного тау-белка в виде нейрофибриллярных клубков, в последнее время пристальное внима-ние уделяется и другим важнейшим процессам на клеточном и молекулярном уровнях, сопровождающим развитие заболевания. В современных исследованиях нейродегенеративных заболеваний все больший интерес вызывает роль митохондрий. Гипотеза митохондриального каскада предполагает, что дисфункция митохондрий играет ключевую роль в прогрессировании нейродегенеративных процессов. Недавние исследова-ния показывают, что клетки имеют способность обмениваться митохондриями между собой. Этот процесс, известный как горизонтальный пере-нос митохондрий, позволяет клеткам обмениваться как здоровыми, так и поврежденными или дисфункциональными митохондриями, перемещая их из одной клетки в другую для дальнейшего восстановления или деградации, что в целом поднимает вопрос о возможности использования в рамках терапии нейродегенеративных заболеваний митохондриальной трансплантации.
Рассмотрены два аспекта: горизонтальный перенос митохондрий и митохондриальная трансплантация. Горизонтальный перенос митохондрий от-крывает новые горизонты в понимании клеточной коммуникации и взаимодействиях. Способы горизонтального переноса митохондрий представ-лены и описаны в деталях. Кроме того, показана актуальность и новаторский характер митохондриальной трансплантации, процедуры, при ко-торой здоровые митохондрии передаются в клетки или органы с дисфункциональными митохондриями. Обсуждены различные методы митохон-дриальной трансплантации и их потенциальное применение в медицине. Представленная информация о новых исследованиях и перспективах в области митохондриальной биологии и терапии расширяет понимание функции и роли митохондрий в живых организмах.
