8 samples of fatty tissue, stored in preservation solution with different formaldehyde concentration and pH value of 18 and 37 ° C, was studied. The purpose was to determine concentration of fatty acids in preservation solution, as well as to study the effect on preserved biological samples, such parameters as concentration of formaldehyde , pH, temperature. To determine the free fatty acids in preservation solutions, sample derivatization was performed in 0,5 % HCl and analyzed by gas-liquid chromatography (GLC). Methyl esters of fatty acids was formed by trans-esterification with methanolic potassium hydroxide, then analyzed by GLC. Experimental data were obtained by analyzing fatty acids in preservative solutions with different parameters, such as pH value of 8 and 9, stored at 18 and 37 ° C, after the 1st and 2nd year of storage, and without formaldehyde and with formaldehyde concen-tration of 0,05% and 0,15%. It was shown that the method can be used to determine fatty acids in preservative solutions. It is proposed to use the results of the study, as the method for determining fatty acids for all solutions in which biological samples are stored. The effectiveness of the technique is determined by using of a small amount of preservative solution (1 ml), reagents, as well as a small amount of time for sample preparation.
