Brief overview on the basic experimental chemotherapeutic and biological studies in Research Institute of Medicinal and Aromatic Plants on natural and synthetic bisbenzylisoquinoline alkaloids (BBIA) of Russian flora, of their derivatives and intermediates, synthetic diphenyl-ether containing analogues of natural BBIA are presented in order to find out biologically active pharmacophore. Prospects of their use in medicine are also discussed. There was studied structural and stereochemical interrelation between BBIA relationships of biological activity with 3D structure of alkaloids, N- and O-substitution in aromatic nucleus and intramolecular connection between groups are shown.
